The FDA has approved Zoryve cream (roflumilast) 0.05% for the treatment of children aged 2 to 5 years with mild to moderate atopic dermatitis.
“Younger children with AD are children with specific medical needs,” Dr. Lawrence F. Eichenfield, chief of pediatric and adolescent dermatology at Rady Children’s Hospital in San Diego and associate chair of the department of dermatology at the University of California, San Diego, told Healio. “We often see a high body surface area, even in some mild cases among children, and there is even greater concern about the side effects of topical steroids. We are pleased to now have this highly effective phosphodiesterase 4 inhibitor, a class of drugs that shows high safety in this population.”
The approval was supported by three studies: the phase 3 INTEGUMENT-PED study, the INTEGUMENT-OLE extension study, and a phase 1 pharmacokinetic study.

I ricercatori hanno osservato un miglioramento del 75% nella gravità della malattia nel 39,4% dei pazienti trattati con Zoryve rispetto al 20% dei pazienti trattati con veicolo (P < 0,0001). Una porzione più ampia del gruppo Zoryve ha anche riscontrato una riduzione di quattro punti nella gravità del prurito rispetto al gruppo veicolo (35,3% vs. 18%; P = 0,0002). I ricercatori hanno osservato che il prurito ha iniziato a diminuire entro le prime 24 ore dall’applicazione di Zoryve crema 0,05% (P = 0,0014).
The safety profile was consistent with the favorable results seen in adult studies. The rate of treatment-related adverse events was low in both groups. Less than 1% of caregivers reported that their child experienced tingling, burning, or discomfort associated with Zoryve cream 0.05%.
30.2% of the 1,247 children and adults enrolled in the 56-week open-label extension study switched to proactive application of Zoryve twice weekly after achieving a clear vIGA-AD score. These participants achieved a median disease control duration of 238 days. This result was consistent with the 281 days observed among adults and children aged 6 years and older also treated with Zoryve twice weekly.
"It's not a steroid, so there were no additional side effects with prolonged use," Eichenfield said, adding, "This extension study showed excellent safety data."
